CRISPR-Cas9 mediated phage therapy as an alternative to antibiotics

Abstract Inappropriate use of antibiotics is globally creating public health hazards associated with antibiotic resistance. Bacteria often acquire resistance by altering their genes through mutation or acquisition plasmid-encoding genes. To treat drug-resistant strains bacteria, the recently developed CRISPR-Cas9 system might be an alternative molecular tool to conventional antibiotics. It disables antibiotic-resistance (plasmids) deactivates bacterial virulence factors and sensitizes bacteria site-specific cleavage crucial domains genome. This uses phages as vehicles for CRISPR-cas9 delivery into bacteria. Since are species-specific natural predators they capable easily injecting DNA target The CRISPR packaged phagemid vectors, in such a way that containing plasmid sequence specific sequences were made targeted. Upon delivery, Cas9 programmed recognize guide RNA thereby causing double-strand targeted loss drug plasmid, which results cell death. Remarkably, safety efficacy this newly biotechnology biocontrol product need further refined its usage clinical translation.
